Human follicle-stimulating hormone (HFSH) labelled with radioactive iodine may be used as a tracer for measurement of metabolic clearance and production rates of HFSH in man if the iodinated hormone is shown to be cleared from the plasma in a similar manner to the endogenous form. Sufficient highly purified HFSH has not been available for clearance studies of the unlabelled hormone in man. Labelled and unlabelled HFSH were simultaneously injected intravenously into monkeys (Macaca mulatta) to compare their metabolic clearance.
HFSH (LER-780, a gift from Dr L. Reichert, Emory University, Atlanta, Georgia) was iodinated with 131I to sp. act. of 50–150 μCi/μg (Greenwood, Hunter & Glover, 1963). It was separated from free 131I by passage through a Sephadex G-75 column.
Six monkeys (three normal, in the luteal phase of the cycle; three ovariectomized—hysterectomized) were anaesthetized with sodium pentobarbitone, 30 mg/kg, i.v., and were given i.v. 1·6 ml
