Pinealocytes constitute 90% of the cell population in the pineal organ (Quay, 1965) and are thought to have an endocrine function (Kelly, 1962; Thieblot & Blaise, 1963; Zweens, 1965). Removal of the pineal in prepubertal rats induces gonadal hypertrophy (Kitay, 1954) and Gittes & Chu (1965) reported that the effects of pinealectomy on the oestrous cycle of female rats could be consistently reversed by transplantation of isogeneic pineal tissue. It seems, therefore, that a pineal—gonadal interrelationship exists. This investigation was undertaken to study the cellular changes in the rat pineal gland after gonadectomy.

It has been suggested that the oestrogen secreted during pro-oestrus plays a role in sensitizing the uterus for implantation of the blastocyst in the mouse. This suggestion was based on indirect evidence using as end points the deciduoma reaction (Finn, 1966) and mitotic activity of the uterus (Finn & Martin, 1970). In contrast, Humphrey (1969), also working with the mouse, furnished direct evidence showing that priming with oestrogen was not necessary for implantation. Finn & Martin (1970) argued that Humphrey was able to obtain implantation because the nidatory oestrogen surge was preceded by 4 days of progesterone treatment, and, had the progesterone treatment been shorter, oestrogen priming would have been necessary for implantation. The aim of the present study was to determine the progesterone priming required, before the nidatory oestrogen surge, for implantation and embryonic development in the rat.

SUMMARY

The binding characteristics of corticosterone by rat liver were studied by a displaceable binding technique. The binding of corticosterone to protein fractionated by gel filtration and density gradient centrifugation has been carried out as a preliminary determination of the nature of the binding sites. The results were analysed and showed three types of binding sites for corticosterone with the characteristic association constants at 0° of K 1 = 1·2 × 1010, K 2 = 1 × 108 and K 3 = 1 × 104 1./mole. Percentage displacement of corticosterone from the nuclear fraction did not differ significantly from that from tissue or the mitochondrial-microsomal fraction. The K 1 and K 2 sites persisted in separated buffer-soluble fractions but were destroyed on mild heating leaving only the K 3 sites.

Abstract

Inhibin and activin are members of the transforming growth factor β (TGFβ) family which can regulate cell proliferation in a number of tissues. The presence of inhibins and the related proteins, activins, in the prostate has been implicated by the detection of activin type II receptors. The aim of this study was to determine whether or not immunoactive (ir) inhibin and ir-activin are present in the rat prostate and to study the acute regulation by androgens. The results showed that mRNAs for the α and β inhibin subunits were detected in rat prostate by reverse transcription-PCR together with ir-inhibin and ir-activin in prostate cytosols. The levels of ir-activin in the prostate (223 ± 44 ng/gland) were greater than the levels of ir-inhibin (6·89 ng/gland), and activin immunoreactivity was localised to the epithelial cells. The presence of these proteins and the subunit mRNAs suggests that these proteins are produced in the prostate and may have a role in prostate function. The study of the effect of androgen withdrawal on the levels of ir-activin and ir-inhibin in these tissues showed no change in the content of ir-inhibin or ir-activin (ng/g tissue) after 3 days of castration or following the administration of the cytotoxic drug ethane dimethane sulphonate (EDS), although there was a significant (P<0·01) decline in prostate weight. Fourteen days after EDS treatment, as the prostate weight fell significantly lower, the amount of ir-inhibin and ir-activin per prostate gland was significantly (P<0·01) reduced although the concentration was unaffected. These data demonstrate, for the first time, that inhibin α and β subunit mRNA and ir-inhibin and ir-activin are present in the prostate; the role of these proteins in prostate function remains to be established.

SUMMARY

In contrast to results in rabbit and dog foetuses, Wolffian duct stabilization in male rat foetuses was inhibited by cyproterone acetate. On the other hand, the stabilizing effect of methyltestosterone in female rat foetuses could be abolished even when excessive androgen doses were given. The hormonal factor(s) involved in normal Wolffian duct stabilization in male rat foetuses remains to be clarified.
