Both heating (Laron, Yed-Lekach, Assa & Kowadlo-Silbergeld, 1964a) and enzyme digestion (Laron, Yed-Lekach, Assa & Kowadlo-Silbergeld, 1964b) alter the immunological properties of human and bovine growth hormone. The purpose of the present investigation was to study the effect of heat and peptic digestion on the immunological properties of sheep prolactin, which has an electrophoretic mobility similar to that of human growth hormone (Laron & Assa, 1963).

Sheep prolactin was supplied by the Endocrinological Study Section of the U.S. National Institute of Health; batches P-S-3 and P-S-4 were used. The preparation of antiserum and the procedures of agar gel double diffusion, electrophoresis and immunoelectrophoresis were performed as previously described (Laron & Assa, 1962). Test tubes containing 2 mg. hormone dissolved in 0·2 ml. water were heated in a water bath at 100° at pH 5·5–6·5.
